Their carbohydrate and surface-charge characteristics were examined with fluorescence, lectin, and electrophoretic methods. The native cartilage model proteoglycan was found to have a higher percentage of highly sulfated glycosaminoglycan than the degraded one, and the sulfate-rich regions showed a decreased tendency to adsorb hyaluronate in both the native and degraded cartilage model proteoglycans. The surface-charge characteristics of the native and degraded cartilage model proteoglycans were quite different, although the net charge of the surface of the native cartilage model proteoglycan was higher than that of the degraded one. The lysine-rich region in the core protein of the native cartilage model proteoglycan was more accessible to the lectin WGA than that of the degraded one, while there was no difference in the accessibility of this region between the native and degraded cartilage model proteoglycans. On the other hand, the presence of chondroitin sulfate reduced the surface-charge properties of the native cartilage model proteoglycan.
